Rapid and automated isolation of high-quality Plant DNA

The extraction of high molecular weight DNA (HMW DNA) from plant tissues is a critical — and often challenging — step in preparing samples for long-read sequencing (Oxford Nanopore, PacBio, etc.).

Following the recent acquisition of the MagnetaPure 32+, we were able to test the NucleoMag® HMW DNA kit on this automated system. In collaboration with Macherey-Nagel, we have written an application note detailing this extraction method and the results obtained.

Thanks to a simple, fast, and automated protocol, up to 32 samples can be processed in just 35 to 45 minutes. An additional purification step has been included to further improve DNA quality for tricky samples.

This method yields DNA fragments over 100 kb in length, meeting the requirements of next-generation long-read sequencing technologies and being compatible with a wide range of plant species, including the most complex ones.
